41 Differential Gene Expression of the Bovine Rumen Epithelial Microbiota During a Sub-acute Ruminal Acidosis (SARA) Challenge

چکیده

Abstract Sub-Acute Ruminal Acidosis (SARA) is a metabolic disorder in dairy cattle characterized by lowered ruminal pH, as result of high grain diets. Dairy cows experiencing SARA show decreased milk production and impaired health; thus, has major economic impacts. been associated with changes the rumen epithelial microbiota. Previously, we performed meta-transcriptome sequencing on biopsy samples from 3 fistulated Holstein before (baseline) after switching to concentrate feed induce analyze gene expression In this previous analysis, 1,607 features were detected using KEGG Ontology reference database, including wide variety genes, well stress response, motility, other functions. However, only differentially expressed between baseline conditions approach. Here, present re-analysis meta-transcriptomics data an improved bioinformatics pipeline reveal additional genetic diversity genes. We de novo transcriptome co-assembly Trinity, mapped reads assembled contigs, used RSEM determine read counts, DESEQ2 for detection genes (Q < 0.05). Features annotated EggNOG, providing taxonomic predictions GO, COG, KEGG-based functional prediction. Using method, total 76,861 transcripts assembled, 4,935 conditions, representing substantial improvement over analysis. Our preliminary analysis indicates levels central metabolism housekeeping oxidative response outer membrane proteins among bacteria. Differentially archaeal primarily upregulated under included involved methanogenesis. Further analyses are ongoing will provide insight into microbiota, their association SARA.

The monitoring, prevention and treatment of sub-acute ruminal acidosis (SARA): a review.

Sub-acute ruminal acidosis (SARA) has become an increasing problem in well-managed, high yielding dairy herds and the monitoring of groups of cows for signs of the condition is now crucial. Rumenocentesis may be ethically questionable but the technique remains the most reliable means of diagnosing SARA. Continuous measurement of ruminal pH may however be possible in the future. Parameters refle...

Impact of Subacute Ruminal Acidosis (SARA) on Ruminal Microbiome, Lipopoly saccharide and Bioamine and Rumen Epithelial Health of Dairy Cows

Maximizing milk yield without impairing host health is a great challenge for dairy industry. However, the high concentrate feeding often unavoidably increases the risk of Subacute ruminal acidosis (SARA) in dairy cows, characterized by periodic episodes of low ruminal pH. The economic costs caused by SARA are enormous, while its pathogenesis is not very clear. As the first and the main digestiv...

Subacute ruminal acidosis (SARA) challenge, ruminal condition and cellular immunity in cattle.

Subacute ruminal acidosis (SARA) is characterized by repeated bouts of low ruminal pH. Cows with SARA often develop complications or other diseases, and associate physiologically with immunosuppression and inflammation. Ruminal free lipopolysaccharide (LPS) increases during SARA and translocates into the blood circulation activating an inflammatory response. Ruminal fermentation and cellular im...

Bovine rumen epithelium undergoes rapid structural adaptations during grain-induced subacute ruminal acidosis.

Alterations in rumen epithelial structure and function during grain-induced subacute ruminal acidosis (SARA) are largely undescribed. In this study, four mature nonlactating dairy cattle were transitioned from a high-forage diet (HF; 0% grain) to a high-grain diet (HG; 65% grain). After feeding the HG diet for 3 wk, the cattle were transitioned back to the original HF diet, which was fed for an...

Effects of a bacterial probiotic on ruminal pH and volatile fatty acids during subacute ruminal acidosis (SARA) in cattle

Effects of a bacterial probiotic (BP) on ruminal fermentation and plasma metabolites were evaluated in four Holstein cattle (body weight, 645 ± 62 kg; mean ± SD) with induced subacute ruminal acidosis (SARA). SARA was induced by feeding a SARA-inducing diet, and thereafter, 20, 50 or 100 g per head of a commercial BP was administered for 7 consecutive days during the morning feeding. Cattle wit...
